Pacific Journal of Health · May 16, 2020: "Phytocannabinoids as therapeutic agents to combat chronic gingival disease." The therapeutic potential of cannabis has been harnessed for thousands of years yet the United States Food and Drug Administration (FDA) has still not approved cannabis as a safe or effective drug. The FDA has, however, approved the manufacturing of pharmaceutical drugs, which contain a “synthetic version of a substance that is present in the marijuana” and other compounds that mimic its action. A search of the US National Library of Medicine and National Institute of Health for keywords “CBD” and “Periodontitis” together yields only 2 publications. Although the current State and Federal regulations on the use of cannabis for research and medicinal purposes seem to contradict one another, there is much room for optimism as the number of States allowing its use increases. This manuscript highlights the recent advances made in cannabinoid research as it pertains to oral health and gingival inflammatory disease. With a brief overview of the endocannabinoid system and its network of receptors and ligands, such as CBD, this manuscript provides the reader with the foundational knowledge necessary to answer common patient questions in a clinical setting.

Cancers Jan 12, 2020 "Super-charged NK cells inhibit poorly differentiated pancreatic tumor growth and metastasis in humanized BLT mice." Patient-derived differentiated pancreatic tumors, or those resected from the pancreas of super-charge NK injected hu-BLT mice exhibiting differentiated phenotype, grew slowly, did not metastasize and were susceptible to chemotherapeutic drugs, whereas those obtained from tumor-bearing mice without NK injection grew fast, metastasized and remained susceptible to NK-mediated cytotoxicity but were resistant to chemo-drugs indicating their undifferentiated/stem-like phenotype. In-vitro NK-differentiated tumors did not grow or metastasize in hu-BLT mice, however, when NK-differentiation of tumors were blocked with antibodies to IFN- and TNF-, tumors grew and metastasized. Much greater percentages of hCD45+ immune cells were recruited to tumors dissociated from NK injected tumor-bearing mice and fed with AJ2 probiotic bacteria. Loss of NK cell function within blood, spleen and bone-marrow in tumor-bearing mice was restored by injection of NKs. Therefore, NK cells prevent tumor establishment by specific recognition, selection and differentiation of cancer stem cells/undifferentiated-tumors in hu-BLT mice, closely resembling human disease.

Masters Thesis · Jan 1, 2017 "Supercharged Natural Killer cells preferentially expand CD8+ Cytotoxic T cells while targeting CD4+ Helper T cells." NK mediated expansion of T-cells preferentially expands CD8+ but not the CD4+ subsets in both healthy and cancer donors. This results in a lower CD4/CD8 ratio. While NK Cells target CD4+ helper T-cells they do not target their CD8+ cytotoxic counterparts. This study also reveals that Cancer patients NK cells have reduced cytotoxic ability and secrete much less IFN-γ than healthy donors and Osteoclast and sAJ2 expanded super- charged NKs intended for immunotherapeutic use from healthy donors expand more immune cells at a faster rate. NK immunotherapy was studied in combination with supplementation of probiotic bacteria in the humanized mouse model. NK immunotherapy increased CD8+ cytotoxic T cell population in immune tissue compartments and Increased immune infiltration in the tumor microenvironment, while probiotic supplementation further enhanced these effects, resulting in decreased tumor burden. This novel expansion technique can provide a non-invasive and highly effective treatment for the treatment of oral squamous cell carcinoma and other tumorigenic cancers.

Cancers · Dec 24, 2019: "Probiotic-Treated Super-Charged NK Cells Efficiently Clear Poorly Differentiated Pancreatic Tumors in Hu-BLT Mice" In this study we determined the role of super-charged NK cells in immune mobilization, lysis, and differentiation of stem-like/undifferentiated tumors implanted in the pancreas of humanized-BLT (hu-BLT) mice fed with or without AJ2 probiotics. The phenotype, growth rate and metastatic potential of pancreatic tumors differentiated by the NK cells (NK-differentiated) or patient derived differentiated or stem-like/undifferentiated pancreatic tumors were investigated.

Cancer Immunity and Immunotherapy · Apr 1, 2017 " Novel strategy to expand super-charged NK cells with significant potential to lyse and differentiate cancer stem cells; Differences in NK expansion and function between healthy and cancer patients" Natural killer (NK) cells are known to target cancer stem cells and undifferentiated tumors. In this paper, we provide a novel strategy for expanding large numbers of super-charged NK cells with significant potential to lyse and differentiate cancer stem cells and demonstrate the differences in the dynamics of NK cell expansion between healthy donors and cancer patients. Decline in cytotoxicity and lower interferon (IFN)-γ secretion by osteoclast (OC)-expanded NK cells from cancer patients correlates with faster expansion of residual contaminating T cells within purified NK cells, whereas healthy donors' OCs continue expanding super-charged NK cells while limiting T cell expansion for up to 60 days. Similar to patient NK cells, NK cells from tumor-bearing BLT-humanized mice promote faster expansion of residual T cells resulting in decreased numbers and function of NK cells, whereas NK cells from mice with no tumor continue expanding NK cells and retain their cytotoxicity. In addition, dendritic cells (DCs) in contrast to OCs are found to promote faster expansion of residual T cells within purified NK cells resulting in the decline in NK cell numbers from healthy individuals. Addition of anti-CD3 mAb inhibits T cell proliferation while enhancing NK cell expansion; however, expanding NK cells have lower cytotoxicity but higher secretion of IFN-γ. Expansion and functional activation of super-charged NK cells by OCs is dependent on interleukin (IL)-12 and IL-15. Thus, in this report, we not only provide a novel strategy to expand super-charged NK cells, but also demonstrate that rapid and sustained expansion of residual T cells within the purified NK cells during expansion with DCs or OCs could be a potential mechanism by which the numbers and function of NK cells decline in cancer patients and in BLT-humanized mice.
